METHODS
Transgenic plant production
-
We perform routine transformations and regeneration of stable transgenic for Arabidopsis thaliana and Medicago truncatula, via Agrobacterium tumefaciens.
-
for Medicago truncatula, the stable transgenic lines are obtained via a regeneration via embryogenesis (Chabaud et al. 1996)
-
For Arabidopsis thaliana transformation we use the the "floral dip" method (Bechtold et al. 1993)
-
Tomato is transformed with Argrobacterium tumefaciens starting with cotyledons.
-
For Brachypodium distachyon, the model monocotyledon plant, the transformation technique was transferred from our colleagues at INRAE Versailles. It consist in the Agrobacterium tumefaciens mediated transformation via somatic embryogenesis.
In developement
-
In collaboration with a team at LIPME, the service developed a quick method for the transient expression of Medicago truncatula roots (Rembliere et al. 2018).
-
The service has developed Medicago truncatula mutant lines that are more embryogenic in order to increase the transformation efficiency.
